Abstract :ÖZET: Bu çalışmada, osteoartrit insert ignore into journalissuearticles values(OA); gelişiminde terapötik hedef olabileceği düşünülen transkripsiyon faktörlerinden NF-κB’nin rolünün belirlenmesi amaçlanmıştır. Çalışmaya primer diz OA tanısı alan 100 hasta ve 50 sağlıklı birey dahil edildi. Periferik kandan mononüklear hücre insert ignore into journalissuearticles values(PKMH); izolasyonu fikol yoğunluk gradyanı yöntemiyle gerçekleştirildi. NF-κB aktivasyonu hücrelerden hazırlanan nüklear ekstraklarda ELISA metodu ile değerlendirildi. Hasta ile kontrol grubu arasında NF-κB aktivitesi değerlendirildiğinde, kontrol grubuna göre bir artış olduğu insert ignore into journalissuearticles values(p=0.405);, cinsiyete göre de artış gösterdiği insert ignore into journalissuearticles values(p=0.260; p=0.610); ancak bu artışların istatistiksel olarak anlamlı olmadığı belirlendi. Bununla birlikte Kellgren Lawrence insert ignore into journalissuearticles values(K-L); evreleme sistemine göre sınıflandırdığımız OA hastalarında NF-κB aktivitesi yönünden evreler arasında istatistiksel bir fark olmadığı insert ignore into journalissuearticles values(p=0.229); bulundu. Ancak NF-κB aktivite değeri kontrol ve diğer evrelerle karşılaştırıldığında erken evrede insert ignore into journalissuearticles values(evre 1); daha yüksek olduğu gözlendi. İnflamatuar sürecin OA patogenezine katkısı olabileceği düşüncesinden yola çıkarak, evrelerine göre sınıflandırdığımız OA hastalarında, NF-κB aktivite değerinin kontrol ve diğer evrelerle karşılaştırıldığında bir fark göstermediği bulundu. In this study, blood samples were collected from 100 patients with a diagnosis of OA and 50 healthy subjects. In these samples peripheral mononuclear blood cells insert ignore into journalissuearticles values(PBMCs); were extracted by standard Ficoll density-gradient centrifugation. NF-κB activation was quantified by way of enzyme-linked immunosorbent assay insert ignore into journalissuearticles values(ELISA); using the NF-κB kit according to the manufacturer`s instructions. When the NF-κB activity is considered between patients and the control group, there is an increase in patients compared to the control group insert ignore into journalissuearticles values(p=0.405);. At the same time,   it has been determined that increased by gender insert ignore into journalissuearticles values(p=0.260; p=0.610);. But these increases were not statistically significant. When we classify OA patients according to Kellgren-Lawrence insert ignore into journalissuearticles values(K-L); grading system, we were not found statistically significant difference between NF-κB activity insert ignore into journalissuearticles values(p=0.229);. But the value of NF-κB activity was significantly higher at early stage insert ignore into journalissuearticles values(grade 1); compared to control and other stages. We are based on, inflammatory processes may contribute to the pathogenesis of OA patients, which were classified according to grade, but were not found difference between NF-κB activities in the patients groups.
