Abstract :Özet Metotreksat insert ignore into journalissuearticles values(MTX);, bazı kanser hastalarının vazgeçilmez ilacıdır. Fakat MTX`in oluşturduğu yan etkiler, doz kısıtlamasına veya tedavinin sonlandırılmasına götürebilir bu da hastanın sağ kalımını azaltır. Bu sebeple MTX`in en önemli yan etkilerinden olan hepatotoksisite ve nefrotoksisitenin oluşum mekanizmasının bilinmesi ve buna karşı önlemlerin alınması oldukça önemlidir. Yapılan birçok çalışma ile MTX`in sebep olduğu hepatotoksisite ve nefrotoksisitede genellikle reaktif oksijen türleri insert ignore into journalissuearticles values(ROT);`nin meydana getirdiği oksidatif hasar sorumlu tutulmuştur. Bu nedenle de oluşacak bu yan etkilerden korunmak için antioksidan ajanlarla birlikte kullanılması gerekliliği ön plana çıkmıştır. Anahtar Kelimeler Antioksidan; Metotreksat; Oksidatif Stres; Reaktif Oksijen Türleri Abstract Methotrexate insert ignore into journalissuearticles values(MTX); is one of the indispensable chemotherapeutic agents for cancer patients. But, MTX induce some adverse effects such as nephrotoxicity and hepatotoxicity. Thus cause dose limitations and terminate the cancer therapy. This will cause decrease in the survival of cancer patients. Hence, we aimed to summarize the MTX`s action mechanism on hepatotoxicity and nephrotoxicity, which of those are most known adverse effects of MTX. However, taking precautions in MTX treatment is crucial in chemotherapy. According to the literatures, reactive oxygen species insert ignore into journalissuearticles values(ROS); triggered oxidative stress is responsible in MTX-induced hepatotoxicity and nephrotoxicity. Hence, the MTX treatment must be accompanied with antioxidants to protect the patients from those adverse effects.
