High Frequency Adventitious Shoot Regeneration From Petiole and Leaf Explants of Endemic Astragalus Polemoniacus Bunge

Abstract :High frequency adventitious shoot regeneration was obtained by using petioles and leaves of endemic Astragalus pole-moniacus Bunge. Various concentrations of 6-benzylaminopurine insert ignore into journalissuearticles values(BAP);, α–naphthaleneacetic acid insert ignore into journalissuearticles values(NAA); and thidiazuron insert ignore into journalissuearticles values(TDZ); were added to Murashige and Skoog insert ignore into journalissuearticles values(MS); medium. The highest frequency of adventitious shoot regeneration insert ignore into journalissuearticles values(100%); and the highest number of adventitious shoots per explant were obtained from petiole explants on a medium containing 4 mg/l BAP and 0.1 mg/l NAA. The shoots developed in vitro were transferred to hormone-free medium or media supplemented with 0.5, 1 and 2 mg/l NAA for rooting. The best root formation was observed in hormone-free or 2 mg/l NAA containing media. The rooted plantlets were acclimatized in pots covered with plastic bags.
