Abstract :ÖZET Geliştirilen bu yöntem selefolJ, .kültür tekniğinin uygun bir boyar/ıa ile birle$tirilip, takiben süreli preparatlarımn /ıazıdanmasıdır. Colletotrıcht!m atramentarium (B. et" Br.) Taub., Rhynchosporium secalis (Oudem.) J.J. Davis fiıngus'ları uygun bi~ besi yeri üzerine yerleştirilen selefon diskler üzerinde gelişmeye bırakıldı. Dllha sf)nra belirli aralıklarla alınan selefon diskler, üzerlerindeki fungus ile blrlikte periodik asit Schiff yöntemi veya analin mavisi ile boyanıp, eti! alkol serisi vexylol den geçirildikten :Sonra Kanada balzamı, Euparal veya Entellan kullanılarak süreli preparatları yapıldı. Bu yöntem fu~gus'ların katı besi yerjnde gösterdikleri gelişmeyi incelemede' vi'" bunların fotoğraflarımn çekilmesinde çok iyi .~o'1;uç }'ermektedir. . Ayrıca süreli preparatlar bir referaJ1s materyali olarak da saklanabilmektedir. SUMMARY A METHOD FOR OBSERVING AND PHOTOGRAPHING THE DEVELOPMENT OF FUNGI ON SOLID MEDIUMS The method deseribed is the eombination of ıl cellophane culture technique witha staiııihg method and then the preparation of permanent mounts. 'The fungi . f Ce/letotrichum atramentarium (B. et BL). Tııub., RhyııchosporiumsecaHs (Oudem.) J.J. Davis ate grown on cellophane discs pIaeed on a suitable medium. Then the cellophane discs with their fungus colony are carefulIy Iifted from the surface of the , medium and stained with Periodic acid Schiff method or analin blue. For the preparatian of permanent mounts theyare passed through graduated series of ethyl alcohol and transfered ·to xYloL. Then theyare mounted: on microscope slides' İn Canada ba1sam. Eupara1 or Entellen. .' This technique perrnit the preparation of permanen-t môuuts r-etaıning diagnostic characteristics of spore stages and mycelium besides giving an opportunity of observing and photographing the development of a fungus on cultiıre mediums.
