Molecular Investigation of Mycoplasma Species in the Lungs of Sheep with Pneumonia

Abstract :This study aimed to investigate the presence of Mycoplasma species in macroscopically detected pneumonia lesions in the lungs of sheep slaughtered in a slaughterhouse in Şanlıurfa using molecular techniques. For this purpose, a total of 100 lung samples were collected from ten different flocks in the Siverek region, each represented by ten lungs with macroscopic lesions. The samples were transported under cold chain conditions to the Laboratory of the Department of Microbiology, Faculty of Veterinary Medicine, Harran University. From each sample evaluated as pneumonic, two serial tissue portions were collected: one for DNA extraction from tissue to be used in direct PCR and the other for examination using the culture-PCR method. DNA was isolated from both tissue samples and cultured media. The DNA samples were tested using species-specific primer pairs targeting M. ovipneumoniae, M. mycoides subsp. capri, M. capricolum subsp. capripneumoniae, M. capricolum subsp. capricolum, M. agalactiae, and M. arginini. Five samples (5%) were found positive for M. mycoides subsp. capri by culture-PCR, while no positive results were obtained for other Mycoplasma species by either direct PCR or culture-PCR. The study revealed that culture-PCR was more advantageous than direct PCR for detecting the causative agent. In conclusion, M. mycoides subsp. capri was identified as an etiological factor in pneumonia cases in the region.
