Study of Adventitious Shoot Formation in Leaves of Grape (Vitis spp.)

Abstract :The effects of genotype, source of explant and nutrient medium on adventitious shoot formation were examined in the leaves of 5 grape cultivars insert ignore into journalissuearticles values(Atasarisi, Çavus, Kalecik Karasi, Sultani çekirdeksiz and Yapincak);, 2 rootstocks insert ignore into journalissuearticles values(Kober 5BB insert ignore into journalissuearticles values( V. berlandieri x V. riparia); and 41 B M.G. insert ignore into journalissuearticles values(Chasselas x V. berlandieri);); by organogenesis. In this study, petioles and laminae isolated from in vitro shoots were used as the sources of explant. The highest rates of adventitious shoot formation were obtained from petiole explants. Leaf explants were cultured on MS- and NN- based regeneration media containing different concentrations of BAP and NAA. It was determined that the most suitable nutrient medium was MS with 2 mg/l BAP for Kalecik Karasi, Sultani çekirdeksiz, Çavus and Kober 5 BB. In this medium, 34.7-56.0% of petioles and 18.7-22.7% of laminae taken from the Kalecik Karasi, Sultani çekirdeksiz, Çavus and Kober 5 BB formed adventitious shoots. It was also observed that NN with 2 mg/l BAP was the most suitable medium for Atasarisi, Yapincak and 41 B M.G. in that 16.0-34.7% of petioles and 8.0-14.7% of laminae from these genotypes cultured in this medium formed adventitious shoots. The level of adventitious shoot formation varied according to genotype. Adventitious shoot formation in petiole explants was greatest in Kalecik Karasi insert ignore into journalissuearticles values(34%); and Sultani çekirdeksiz insert ignore into journalissuearticles values(32.7%);; moderate in Kober 5 BB insert ignore into journalissuearticles values(23%);, Çavus insert ignore into journalissuearticles values(21.5%); and Yapincak insert ignore into journalissuearticles values(21.2%);; and lowest in Atasarisi insert ignore into journalissuearticles values(18%); and 41 B M.G. insert ignore into journalissuearticles values(8.5%);.
