Abstract :ÖZET Yaprak saydamlaştırma ve fungus boyama metodu özellikle Rhyndwsporium secalis (Oudem) JI Davis'in arpa bitkisi dokularındaki gelişmesini izlemek üzere geliştirilmiştir. Bu metodda enfekteli yapraklar önce laktofenol +metanol+ kloroform karışımında saydamlaştmhp takiben laktofenol-analin mavisi içinde kaynatılarak dokulardaki jimgus boyanmaktadır. SUMMARY A Rapid Leaf elearing and Fungus Staining Technique to Study Host-Pathogen Relationships This leaf clearing and fungus staining tchnique is especially modified to study the development of Rhynchosporiutn secalis (Oudem) J.J. Davis in the tissues of barley plants. in this method infected leaf sections are eleared in a mıxture of lactophenol + metlıanol + clıoloroform at 50°C for about 1 hour. After c1earing, leaf sections are brought to boiling for about 2-3 minutes in lactophenol-analin blue and then rinsed iri lactophenal. Afterwards Ieaf sections are mounted on mic· roscope slides in lactophenol for observation.
